Effect of eutectic formulation on photodegradation kinetics of ornidazole in aqueous state: predicted interaction and molecular binding in docking studies

Abstract: The sensitivity of ornidazole towards UV light is one of the most crucial causes in the significant reduction of the therapeutic efficacy. The effect of eutectic–excipient on photodegradation of ornidazole in aqueous state under the exposure of UV light has been studied. Ornidazole–eutectics were prepared using benzoic acid, citric acid, and malic acid as the former molecule (all at 1:1 molar ratio). Kinetics of ornidazole degradation has been studied in the aqueous state in the presence of the eutectic former under the influence of UV light. The patterned photodegradation has also been compared in the presence of HPMC (1:1 weight ratio). A significant decrease in the first-order kinetics rate constant (K) was observed for the ornidazole–eutectic mixture as compared to the ornidazole alone in the aqueous state. FTIR, DSC, XRD, and SEM study of the formulated sample suggested in support of eutectic formation. The negative energy in the in silico docking score of the eutectic complex indicated additional suggestion towards the stable binding (− 1.5 to − 2.1 kcal/mol).
