Comparison of the photometric evaluation of chromatograms in reflected and transmitted light

Abstract: The accuracy of photometric methods for the evaluation of chromatograms directly from the paper and from a negative in reflected and transmitted light was studied. For measurements a recording microphotometer, a densitometer, and a photometer were used. If 2 filters are used, the quality of the paper has a small effect on the zero value in the recorded data. Measurements were carried out on chromatographic mixts. of amino acids and sugars. The recorded data were evaluated according to the height and area and statistically. In most cases better results were obtained by measuring the height of the waves. In direct evaluation by transmitted light, the relative standard deviation was 1.6-4.2%, for other methods 2.6-5.3%. In transmitted light the sensitivity was increased 5-10 times.
