Oscillographic polarography of nucleic acids

Abstract: Nucleic acid bases (adenine, guanine, cytosine, 5-methylcytosine, thymine, uracil) and purine derivs. (hypoxanthine, xanthine, and uric acid) were investigated by a oscillopolarographic method with a.c. in the medium of M H2SO4, HCOONH4, KCl, and NaOH. All these compds. give inflections on the curves dE/dt = F1(E) and the majority of them and the nucleosides and the nucleotides can be detd. alone or in the mixt. in the concn. 10-5M-10-6M. A highly polymerized deoxyribonucleic acid (I), prepd. from calf thymus or rat spleen, gives characteristic inflections in HCOONH4, NaCl, NaOH, Brdicka's CoIII and NiII solns. In the CoIII and NiII solns., both protein and I can be detd. even in a great excess of I. The anodic inflection in HCOONH4 is caused by the deoxyguanylic acid bound in the I mol. A hypothesis of the structure of I is given.
