Determination of capsaicin. I. Reaction with p-diazobenzenesulfonic acid

Abstract: The coupling of p-diazobenzenesulfonic acid (I) with capsaicin (II) (active component of sharp paprica) is described. I was in a 0.1N HCl soln. and standardized by the extinction of the color developed by coupling with PhOH. In the detn. of II in borate buffer solns. the extinction coeffs. are considerably higher than in glycine buffer solns. at the same pH. The presence of EtOH decreases the sensitivity of the reaction and it was removed by the distn. in an atm. of N. The value of extinction coeff. is const. at borate buffer solns. with pH range 10.68-11.70. By using Thiele's borate buffer, with pH 11.0, the results can be exactly reproduced. The equation for the calibration line is m = 0.03058c - 0.0039.
