Hemoglobin. V. N-terminal amino acids and the number of polypeptide chains in rat hemoglobin

Abstract: Rat hemoglobin terminates in 2 α-polypeptidic chains contg. valine and in 2β-polypeptidic chains contg. methionine. From an acid hydrolyzate of dinitrophenylated globin a valylleucyl sequence was isolated which represents the α-polypeptidic chain in hemoglobin. By stepwise degradation with phenylthiohydantoin (I) leucine and glycine were detd. as N-terminal amino acids. Two α-polypeptidic chains are terminated with a valylleucyl sequence and 2β-polypeptidic chains with a methionylglycyl sequence. Quant. detn. of I derivs. with paper chromatography and a modified method of crystn. of hemoglobin are described.
